Knight hit 62 not out, and Matthews 51 as Barbados posted 144 for 4 from their 20 overs in their Group A match at Edgbaston.
Pakistan’s Nida Dar hit 50 not out from 31 balls, but Pakistan could only muster 129 for 6 in their reply.
Barbados, with seven West Indies players in their squad, next play Australia on Wednesday from 11am TT Time.
Barbados qualified for the Commonwealth Games by winning the T20 Blaze 2019, the West Indies Cricket Women’s T20 Championship.
